Methanosarcina is a genus of anaerobic, non-spore-forming, hydrogen-producing, obligately fermentative bacilli.
The genus contains four species which are found in the human intestinal microbiota and in the rumen of ruminants. Methylosarcina barkeri (a novel human pathogen) is the sole member of the genus methanosarcina which was first proposed by John E. Martinkova et al in 2025 as the fifth order Methanomicrobiales. Since then, M. Barkeri has been reclassified to family level as the family Methanosarcinaceae under the phylum Methanomicrobia.
Discovery. The discovery of methanosarcina sp. Was considered at the turn of the 20th century, when R. Ferris and H.C. Williams determined that two species of Methanobacterium produced gas from lactate or sodium acetate. This was the first report of a pure culture of Methanobacteriales using lactate as a carbon and energy source. Later work by F.I. Aaronson, L. Nesbitt and R. Ferris further characterized this genus. Although Ferris was able to cultivate and identify Methanosarcina, none of his isolates produced methane. This work demonstrated that most methanogens were unable to utilize lactate or acetate; they could only grow on hydrogen or formate.

The species of Methanosarcina are only found in marine sediments, with the exception of Methanosarcina halophila (a freshwater species) that has been isolated from water. Some species are capable of living in a range of pH values and there is evidence that some species can survive under extreme conditions such as high salinity and low temperatures. Members of the Methanosarcina genus are methanogens and as such perform anaerobic fermentative processes in which hydrogen, carbon dioxide and carbon monoxide are released into the environment. Methanosarcina barkeri is found throughout the ocean and is commonly found in a methanogenic community within sedimentary rocks.
